5、Research background：
HER2 is a 185kDa transmembrane glycoprotein that belongs to the human epidermal growth factor receptor family (ErbB family). It is an important biomarker for targeted therapy in breast cancer, gastric cancer and many other solid tumors. It includes three domains: extracellular ligand binding domain (ECD), short transmembrane region and intracellular tyrosine kinase domain. The ECD of HER2 consists of four domains (I-IV). Domain II and IV participate in homologous and heterologous dimerization, while Domain I and III have the ability to bind to ligands. HER2 lacks specific ligands and is the only one that can form dimers with other members, leading to phosphorylation of tyrosine kinase residues in the cytoplasmic domain, thereby activating signaling pathways such as phosphatidylinositol triphosphate kinase (PI3K) and mitogen-activated protein kinase (MAPK), thereby triggering receptors that regulate cell proliferation and survival signaling pathways. This monoclonal antibody is trastuzumab, which is a Her2 antagonist. It binds to the fourth functional domain of the HER2 protein outside the cell membrane, thereby blocking the growth of tumor cells, while mediating cellular immunity and killing tumor cells.
